Selective Autophagy Controls Innate Immune Response Through a Tak1/Tab2/Sh3px1 Axis

چکیده
Selective autophagy is a catabolic route which turns over specific cellular material for degradation by lysosomes, and whose role in the regulation of innate immunity largely unexplored. Here, we show that apical kinase Drosophila immune deficiency (Imd) pathway Tak1, as well its co-activator Tab2, are both novel selective substrates interact with protein Atg8a. We also Atg8a-interacting Sh3px1 downregulation Imd pathway, facilitating targeting Tak1/Tab2 complex to platform through interaction Tab2. Our findings Tak1/Tab2/Sh3px1 interactions Atg8a mediate autophagic complex, turn prevents constitutive activation Drosophila. This study provides mechanistic insight on responses autophagy.

NK-DC crosstalk controls the autopathogenic Th17 response through an innate IFN-γ–IL-27 axis
IFN-γ is a pathogenic cytokine involved in inflammation. Paradoxically, its deficiency exacerbates experimental autoimmune encephalomyelitis, uveitis, and arthritis. Here, we demonstrate using IFN-γ(-/-) mice repleted with IFN-γ +/+: NK cells that innate production of IFN-γ from NK cells is necessary and sufficient to trigger an endogenous regulatory circuit that limits autoimmunity. After immu...
متن کاملHepatitis C Virus-Induced Autophagy and Host Innate Immune Response
Autophagy is a catabolic process that is important for maintaining cellular homeostasis. This pathway in hepatocytes is stimulated and controlled by the hepatitis C virus (HCV)-upon infection-to promote its own replication. HCV induces autophagy indirectly and directly through different mechanisms and temporally controls the autophagic flux. This enables the virus to maximize its replication an...
متن کاملCD38 controls the innate immune response against Listeria monocytogenes.
CD38, adenosine-5'-diphosphate-ribosyl cyclase 1, is a multifunctional enzyme, expressed on a wide variety of cell types. CD38 has been assigned diverse functions, including generation of calcium-mobilizing metabolites, cell activation, and chemotaxis. Using a murine Listeria monocytogenes infection model, we found that CD38 knockout (KO) mice were highly susceptible to infection. Enhanced susc...
متن کاملAutophagy as an Innate Immune Modulator
Autophagy is a fundamental cellular process in eukaryotic cells for maintaining homeostasis by degrading cellular proteins and organelles. Recently, the roles of autophagy have been expanded to immune systems, which in turn modulate innate immune responses. More specifically, autophagy acts as a direct effector for protection against pathogens, as well as a modulator of pathogen recognition and...
